The Louis Armstrong song, originally released in 1967, sees Katie Melua sing with her musical idol, who died of cancer in 1996. Profits from the single go to the British Red Cross.
Following behind Katie Melua and Leona Lewis is Soulja Boy, whose Tell 'Em Crank That single climbed to number three ahead of Mariah Carey's All I Want For Christmas Is You at number four and Girls Aloud at five with Call The Shots.
The Christmas number one single will be announced next Sunday with favourites including X Factor winner, Leon Jackson with When You Believe, Leona Lewis and Katie Melua.
The highest new entry in the UK singles chart came from Michael Buble whose Lost single entered the chart at number 19 ahead of Shaun The Sheep's Life's A Treat festive single. 12 of the top 40 singles are taken by Christmas tracks, including John Lennon's Happy Christmas (War Is Over), Chris Rea's Driving Home For Christmas plus The Pogues, Slade, Wham!, Shakin' Stevens and Band Aid.
Meanwhile, in the album chart, Leona retained her lead at the top of the charts for a fifth week with her debut album, Spirit. Led Zeppelin's Best Of album, Mothership climbed to number four, while The Spice Girls' Greatest Hits album fell from number 13 to 22.
Led Zeppelin, who also staged a live comeback this week, fared better - with best of collection Mothership rising four places to number four.
